To me, it also looks like the mark of the Crucible (we can easily see it on the crucible sets and both crucible knight variants). The colorarion of this specific rune is not golden, but has the specific reddish hue that is mentioned to also be connected to the Crucible (mentioned on the Godrick knight greatshield, which also has a similar color).
The rune itself could be a reference to how the elden beast corrupted the Crucible, creating the erdtree, which is also the source of the runes.
We also know for a fact that the power of the divine bloodline (Marika and her offspring) becomes diluted and weaker as the generations go by (Godrick being the perfect example). As such, it could be possible that the lesser runes we find (even the basic, 200 ones) are newer, while the most powerful ones are older.
My theory is, that this is one of the very first runes created by the elden beast as it was corrupting the Crucible, which explains why it is extremely potent and how it has the exact same shape and hue of the mark of the Crucible, since it wasn't completely reshaped into the Erdtree yet. This also could explain why the lesser runes tend to get smaller, and more golden the "newer" they are. Another prime example of this is the Hero's rune, which has a hue that is less reddish, but still somewhat similar to this one.

If you have PS5 and PS4 and bought the digital edition, you can download the game on both. Then create a second account with PlayStation Plus and you can duplicate the rune to its max, always making a backup save, dropping the runes and then reloading the backup :)

Anyone else notice that the greater the runes get, the more the runes look like an eye?

You can find one on the way to fight the lord of the blood.

Another one is in Mogwyn Palace zone, exploring the cave before the elevator that leads to the boss. Actually You can get this pretty early in the game, just by unlocking Liurnia and doing Varre quest.

You can get one of these very early at Mohgwynn's Palace if you do the Vierre quest.
